Amaç: Hücre içi kalsiyum (Ca2
+) sinyali; hücre proliferasyonu ve farklılaşması, gen transkripsiyonu, apoptozis
gibi birçok hücresel süreçte rol oynar. Yapılan çalışmalarda Ca2
+ sinyalinin kanser ile ilişkili olduğu gösterilmiştir. Transient receptor potential vanilloid 4 (TRPV4) kanalları hücre zarında bulunan Ca2
+ geçirgen
non-selekif katyon kanalıdır. TRPV4 kanalları vücutta birçok dokuda yaygın olarak eksprese edilmektedir.
Hücre içi Ca2
+ miktarının düzenlenmesinde etkisi olan TRPV4 kanallarının kanser hücrelerinin proliferasyonu, apoptozis, migrasyonu ve tümor anjiogenezi ile ilgili önemli rolleri olduğu görülmüştür. Biz bu çalışmada
TRPV4 antagonisti olan RN 1734’ün, insan prostat (PC-3) ve insan meme (MCF-7) kanseri hücre hatlarında
hücre canlılığı ve DNA hasarı üzerine etkilerini araştırdık.
Gereç ve Yöntemler: Çalışmamızda insan prostat (PC-3) ve insan meme (MCF-7) kanseri hücre hatları kullanıldı. RN 1734 1, 5, 25, 50 ve 100 μM’lik konsantrasyonlarda, 24 saatlik PC-3 ve MCF-7 hücre canlılığına olan
etkisi 3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazolyum bromid (MTT) yöntemiyle ölçüldü. RN 1734’ün DNA
hasarına etkisi Comet yöntemine göre belirlendi.
Bulgular: 24 saat süreyle RN 1734’ün özellikle 50 ve 100 μM’lik konsantrasyonlarıyla inkübe edilen PC-3 ve
MCF-7 hücrelerde canlılık düzeyi önemli ölçüde azaldı (p<0.05). Ayrıca 100 μM’lik konsantrasyonda RN 1734
uygulaması sonrası hücre DNA hasarı düzeyinde anlamlı artışın olduğu görüldü (p<0.05).
Sonuç: Biz bu çalışmada TRPV4 antagonisti olan RN 1734'ün, PC-3 ve MCF-7 hücre hatlarında hücre canlılığını azalttığını ve DNA hasarına neden olduğunu bulduk. Bizim bulgularımız, TRPV4 antagonisti uygulanmasının kanser hücrelerine karşı yeni bir tedavi yaklaşımı olabileceğini düşündürmektedir.; Aim: Intracellular calcium (Ca2+) signal pathway has a role in cell proliferation and differentiation, gene transcription and apoptosis. Studies have shown that Ca2+ signal is related to cancer. Transient receptor potential vanilloid 4 channels are Ca2+ permeable non-selective cation channels in the cell membrane. TRPV4 channels are commonly expressed in many tissues in the body. TRPV4 channels, which have an effect on the regulation of intracellular Ca2+ levels, have been shown to have important roles in the proliferation, apoptosis, migration and tumor angiogenesis of cancer cells. In this study, we investigated the effects of RN 1734, TRPV4 antagonist, on cell viability and DNA damage in human prostate (PC-3) and human breast (MCF-7) cancer cell lines. Materials and Methods: Human prostate (PC-3) and human breast (MCF-7) cancer cell lines were used in our study. The effect of RN 1734 on 1, 5, 25, 50 and 100 µM concentrations on 24-hour PC-3 and MCF-7 cell viability was determined by 3- (4,5-dimethylthiazol-2-yl) -2,5- diphenyltetrazolium bromide (MTT) method. The effect of RN 1734 on DNA damage was determined according to Comet Assay method. Results: The viability of PC-3 and MCF-7 cells, which were incubated for 24 hours with RN 1734, especially at concentrations of 50 and 100 µM, was significantly reduced (p<0.05). In addition, there was a significant increase in cell DNA damage level after RN 1734 100 µM concentration application (p <0.05). Conclusions: In this study, we found that RN 1734, TRPV4 antagonist, reduces cell viability and causes DNA damage in PC-3 and MCF-7 cell lines. Our findings suggest that administration of TRPV4 antagonist may be a new therapeutic approach to cancer cells.

Borun topraktaki eksikliği veya yüksek konsantrasyonda bulunması önemli bir abiyotik stres faktörü olabilmektedir. Arabidopsis thaliana’da Cu/ZnSOD (CSD1, CDS2, CSD3), FeSOD (FSD1, FSD2, FSD3) ve MnSOD (MSD1) genlerinden oluşan 7 farklı süperoksit dismutaz (SOD) izoformu tanımlanmıştır. Yapılan çalışmada amaç bor (B) eksikliği veya toksisite koşulları altında Arabidopsis thaliana (L.)’nın yaprak ve kök dokusunda antioksidan sistemin anahtar enzimi olan süperoksit dismutazı kodlayan genlerin ifade düzeylerini Real-Time PCR ile belirlenmesidir. Arabidopsis bitkileri altı hafta hidroponik kültür sisteminde yetiştirilmiş ve 48 saat süreyle borik asit (BA) içermeyen (0 µM) veya yüksek konsantrasyonda BA (3000 µM) içeren besin ortamlarında inkübe edilmiştir. B stres uygulamaları CSD1 ve FSD3 genlerinin ekspresyonunu yaprak dokusunda arttırmıştır. MSD1 geninin mRNA seviyesi yaprak dokusunda B toksisite uygulaması sonucu artarken, kök dokusunda B eksikliğinde artış göstermiştir. B eksikliği ve toksisitesi uygulamaları CSD2, FSD1 ve FSD2 genlerinin ekspresyon düzeyini dokuya bağlı olarak değiştirmiştir. Sonuç olarak, B stresinin kök ve yapraklarda farklı hücresel kompartımanlarda antioksidatif savunmayı tetiklediği ortaya koyulmuştur.; Deficiency or excess of boron in the soil may be an important abiotic stress factor. Seven different superoxide dismutase (SOD) isoforms including Cu/ZnSOD (CSD1, CDS2, CSD3), FeSOD (FSD1, FSD2, FSD3), and MnSOD (MSD1) genes have been identified in Arabidopsis thaliana. The aim of this study is to determine the expression levels of the genes encoding superoxide dismutase, which is the key enzyme of the antioxidant system, in leaf and root tissues of Arabidopsis thaliana (L.) under boron (B) deficiency or toxicity conditions using Real-Time PCR. Arabidopsis plants were grown in hydroponic culture system for six weeks and incubated for 48 hours in mediums excluding boric acid (BA) (0 μM) or including high concentration of BA (3000 μM). B stress tretaments increased the expression levels of the CSD1 and FSD3 genes in leaf tissue. mRNA level of MSD1 gene is increased in result of B toxicity application in leaf tissue, while B deficiency application increased the mRNA level in root tissue. B deficiency and toxicity treatments altered the expression levels of the CSD2, FSD1, and FSD2 genes depending on the tissue. In conclusion, it has been revealed that B stress triggers antioxidative defense mechanisms in different cellular compartments in roots and leaves.

Antioxidant, antimicrobial and anticancer activities of the aspergillin pz and terphenyllin secondary metabolites: an in vitro study

The importance of secondary metabolites obtained from fungi is increasing day by day in terms of human health. Many physiological and pharmacological effects, mainly antimicrobial, anticancer and antioxidant properties of these compounds have been reported. Aim of this study is to determine the anticancer, antioxidant and antimicrobial effects of aspergillin PZ and terphenyllin compounds, which are isolated from Aspergillus and also subjected to limited number of studies. The antimicrobial activity of aspergillin PZ and terphenyllin compounds was determined by using disc diffusion method using different bacteria. The antioxidant property of the compounds was determined by measuring the level of DPPH free radical scavenging. Cytotoxic activity was determined by experiments on human prostate cancer cell lines (PC3 and LNCaP) and over cancer cell line (A2780). Both compounds showed low antimicrobial activity on test bacteria (approximately 2-3 mm zone). High concentrations of applied compounds showed apparent DPPH free radical scavenging activity, while % scavenging activity was quite low at low concentrations. Both compounds showed significant anticancer activity on cancer cell lines (p <0.05). Our results suggest that these two compounds have important biological properties due to their antioxidant and anticancer activities.

Irisin is thought an anti obesity hormone, which regulates body weight and metabolism, including insulin resistance. It is known that obesity is a risk factor in the development of cancer. This study was designed to evaluate whether there is a role of irisin on viability of human prostate cancer cells. In the present study, 0.1, 1, 10 and 100 nM concentrations of irisin were applied to human prostate cancer cells with androgen receptor positive (LNCaP) and androgen receptor negative (DU-145, PC3). Effects of irisin were determined using 3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. At the end of study, all concentrations of irisin reduced viability in the three types of prostate cells, but only 10 and 100 nM concentrations of the irisin caused a significant decreases (p<0.05). Consequently, high concentrations of irisin in both androgen receptor positive and androgen receptor negative cell lines reduced cell viability. These results show that the cytotoxic effects of irisin on prostate cancer cells are not dependent on androgen receptor mechanism.
